So, I've seen lately that a lot of members have gotten some pretty decent gains from E-85 tunes. In my continued search for the best naturally aspirated build, I have been wondering if the higher octane rating of E-85 would allow me to up my compression? If so, how much? 12:1? 12.5:1? 13:1?

I'd go 13:1 all day NA on E-85.

On a street car I probably wouldn't. E85 will definitely let you safely run higher comp with good tuning. I would talk with a seasoned engine builder and/or tuner to see what they feel comfortable with compression wise.

Would you go any higher?
I wouldn't with e85's 104 octane. If it's an NA build there probably wont be a lot of cylinder bleed off like a power adder application which will bring the dynamic compression down. When I build engines for nitrous, I get as close to 16:1 as I can. After calculating the dynamic, I'm usually around 13-13.5:1 ratio which depends on the camshaft selected.

Any updates on this? Where do you find these 13:1 pistons? I am doing a all out NA build with ported GT350 heads either a slightly bored Gen 2 coyote or just a Gen 3 with the CJ and cams combo
