Hoping to save some time looking for a good ground connection for my amplifiers. They will be trunk mounted in a 2015 Convertible. There is a ground wire connected to the very back panel under the plastic trim, but I wonder if there is a better one that others have used.

The reason the spare tire area is chosen is that the battery - is the actual GND in this instance. Theoretically, the best ground would be there, but since that terminal is bonded to the chassis of the vehicle, the spare tire area and the major single structure around it is the best option. Do not connect to jumpers, or other body panels even if they are metal and technically attached, as the resistance to ground could then be exceedingly high.

What about the post that is intended to lock down the spare (if there was one), it has some threaded holes already.

That would work. I would sand the coating off first, so that the ring terminal on your ground wire makes contact with bare metal. Secure the ground wire with a flange head bolt.
