I'm most certainly not answering this for Livernois, but if it is using the same strategy thats in the pcm and also unlocked in the F150, then just a few minutes. So just don't go cut a 1.5 60' from the fuel pump.How quickly/progressively does the tune adjust? For example, say I'm a the bottom of a tank of 93, and fill with e85 a few miles away from the track for a net of ~e80. Is it going to be 'calibrated' by the time of my first run?
Also, does this assume a floor of 93 or does it allow for switching up between 87, 91, 92, 93, etc?

There is an assumed floor of at least 91 octane. So there is no way that you can go from 87 octane or lower and goto E85.How quickly/progressively does the tune adjust?
